要 約
乳牛に消化管運動を賦活および調整するシサプリドを臨床応用する際の基礎データを得るため，健康牛5頭を用いてこの薬物の薬物動態学的解析を実施した．シサプリド（0.1mg/kg）を頸静脈から単回投与し，投与10，20，30，40，50，60，80，100，120，150，180，240，300および360分後に血清を採取した．血清シサプリド濃度はHPLC法により測定し，2コンパートメント・モデルで解析した．その結果，消失相の半減期は1.90±0.18（平均値±標準偏差）時間，総クリアランスは23.17±1.05ml/kg/分，定常状態における分布容は2.95±0.45l/kg，そして平均滞留時間は2.11±0.29時間であった．

Pharmaco-kinetics of Cisapride in Dairy Cattle after Intravenous Administration

SUMMARY
A pharmaco-kinetic study of cisapride in cows was made to provide a basis for the clinical
use of the drug. Cisapride (0.1 mg/kg) was intravenously administered to 5 clinically healthy cows; and
serum was taken at 10, 20, 30, 40, 50, 60, 80, 100, 120, 150, 180, 240, 300, and 360 min after administration.
Serum cisapride concentrations were measured by means of the HPLC method, and pharmaco-kinetic parameters
were analyzed on the basis of a 2hcompartment model. Serum half-life was 1.90±0.18 (mean±SD) hr; total
body clearance was 23.17±1.05 ml/kg/min; volume of distribution at a steady rate was 2.95±0.45 l/kg;
and residence time was 2.11±0.29 hr.
