片腎摘出と他方の腎動脈部分結紮により作成した腎機能低下状態の犬において,作成前(正常腎期間)と作製後(1/4腎期間)に,肝腎排泄性のアンジオテンシン変換酵素阻害薬であるベナゼプリルの常用量 (0.5mg/kg)と20倍量(10mg/kg)を1日1回,15日間経口投与した.正常腎期間には,両投薬量
とも異常な症状と検査所見を認めなかった.1/4腎作成後,腎血漿流量と糸球体濾過量 は約1/3に減少し,BUN濃度と血漿クレアチニン濃度は上昇した.1/4腎期間においても,0.5mg/kgおよび10mg/kg投薬群とも投薬と関係する異常症状は認められず,腎血漿流量
,糸球体濾過量,BUN濃度,血漿クレアチニン濃度等は変化しなかった.

Effects of High Doses of the Angiotensin-converting Enzyme Inhibitor Benazepril
on Dogs with Experimental Kidney Impairment

SUMMARY
The angiotensin-converting enzyme inhibitor, benazepril was orally administered
to dogs for a period 15 days before and after renal ablation. Renal ablation by right-side nephrectomy
and ligation of one branch of the left renal artery were performed in the animals to impair renal function.
One group of dogs received a standard dose (0.5 mg/kg) of benazepril; a second group received 20 times
the standard dose (10 mg/kg). During the period before surgery, neither the standard nor the increased
doses produced remarkable changed in clinical signs and laboratory test results. In 11/2nephrectomized
dogs, renal plasma flow and glomerular filtration rate decreased to approximately 1/3 of what they had
been before surgery, and BUN and plasma creatinine concentrations increased. These variables remained
unchanged in both dose groups during the administration period also in 11/2 nephrectomized
dogs.
