豚の筋肉および腎臓に残留したオキソリン酸,オフロキサシン,エンロフロキサシン,ダノフロキサシンを選択的イオン検出(SIM)法を用いたガスクロマトグラフィー/質量 分析法(GC/MS)で分析する方法を開発した.キノロン剤は,試料から0.3%メタリン酸―アセトニトリル(6:4)で抽出し,Sep-Pak
PS-2で精製後,メタノールで溶解した.精製したキノロン剤は,水素化ホウ素ナトリウムにより還元し,0.1N塩酸を加え,水素原子を付加した.キノロン剤誘導体は,反応溶液からSep-Pak PS-2で抽出後,各薬剤につき3イオンをモニターしたSIM法によりGC/MSで分析した.回収率は筋肉で80.8〜90.5%,腎臓で82.3〜88.6%であり,検出限界値は,ダノフロキサシンが0.02μ

Confirmation of Quinolone Antibacterials in Porcine Meat and Kidneys by Gas Chromatography/Mass Spectrometry Using Selected Ion MonitoringSeiji ASAMI, Kazumi HATSUGAI, Kazuyuki OSAWA,Yoshio NAKABAYASHI and Takashi NAKAJIMA
SUMMARY A gas chromatographic/mass spectrometric method (GC/MS) using selected ion monitoring (SIM) was
developed for the determination of oxolinic acid, ofloxacin, enrofloxacin, and danafloxacin in porcine
meat and kidneys. Quinolone antibacterials were extracted with 0.3% metaphosphoric acid-acetonitrile
(6 : 4) followed by a Sep-Pak PS-2 cleanup procedure. After evaporation, the residue was dissolved in
methanol and reduced with sodium tetrahydroborate treated with 0.1N HCl. Quinolone derivatives were extracted
by a Sep-Pak PS-2 from the reaction mixture and analyzed by GC/MS (SIM). Average recoveries of the drugs
from tissues were in the ranges of 80.8〜90.5% in meat and 82.3〜88.6% in kidneys. Detection limits were
0.01μg/g for oxo- linic acid, ofloxacin, and enrofloxacin and 0.02μg/g for danofloxacin. --------------------J. Jpn. Vet. Med. Assoc., 53, 225〜231(2000) |
